Crotonobetaine reductase from Escherichia coli is composed of two proteins (component I (CI) and component II (CII)). CI has been purified to electrophoretic homogeneity from a cell-free extract of E. coli O44 K74. The purified protein shows l(-)-carnitine dehydratase activity and its N-terminal amino acid sequence is identical to the caiB gene product from E. coli O44 K74. The relative molecular mass of CI has been determined to be 86100. It is composed of two identical subunits with a molecular mass of 42600. The isoelectric point of CI was found to be 4.3. CII was purified from an overexpression strain in one step by ion exchange chromatography on Fractogel EMD TMAE 650(S). The N-terminal amino acid sequence of CII shows absolute identity with the N-terminal sequence of the caiA gene product, i.e. of the postulated crotonobetaine reductase. The relative molecular mass of the protein is 164400 and it is composed of four identical subunits of molecular mass 41500. The isoelectric point of CII is 5.6. CII contains non-covalently bound FAD in a molar ratio of 1:1. In the crotonobetaine reductase reaction one dimer of CI associates with one tetramer of CII. A still unknown low-molecular-mass effector described for the l(-)-carnitine dehydratase is also necessary for crotonobetaine reductase activity. The indirect, non-lethal results of predation, such as reduction in feeding time or restraint in seeking sexual partners and/or natural resources, have a drastic effect on prey populations. In this study, we investigated the behaviour of two serpa tetras Hyphessobrycon eques groups, one wild and the other born and raised in captivity, to evaluate how their feeding behaviour is affected by the avian predator, the rufescent tiger-heron Trigrisoma lineatum (using a taxidermy specimen). For a total of 133 observation hours, the feeding behaviour of each fish group was observed according to the absence (control) or presence (treatments) of a predator near the aquarium surface. The results showed that the presence of a predator on the surface inhibits the feeding behaviour of H. eques. The differences observed between the groups are probably due to the fishes experiences with predators. Our results suggest that fish-eating birds may affect prey populations in streams and ponds perhaps more through non-lethal effects, on feeding behaviour for example, than directly through death rates. 相似文献

Arthrospira (Spirulina) platensis (Nordstedt) Gomont was cultivated under light‐limited conditions in 5‐L open tanks by daily supplying NH4Cl as nitrogen source. Exponentially increasing feeding rates were adopted to prevent ammonia toxicity. The total feeding time (T) was varied between 12 and 20 days, and the starting (m0) and total (mT) quantities of the nitrogen source per unit reactor volume were varied in the ranges 0.19–1.7 mM and 2.3–23.1 mM, respectively. This intermittent addition of the nitrogen source prevented ammonia from reaching inhibitory levels and ensured final cell concentrations (Xm) and cell productivities (Px) comparable with those of batch runs with KNO3. Moreover, the lower nitrogen addition due to the use of NH4Cl rather than KNO3 allowed for higher nitrogen‐to‐cell conversions (Yx/n). These results were evaluated using three‐factor, five‐level, central composite experimental planning, combined with the response surface methodology, selecting T, m0, and mT as the independent variables and Xm, Px, and Yx/n as the response variables. This approach allowed us to identify, through the simultaneous optimization of the variables, T=16 days, m0=1.7 mM, and mT=21.5 mM as the best conditions for A. platensis cultivation at 72 μmol photons·m?2·s?1. Under these conditions, a maximum cell concentration of 1239 mg ·L?1 was obtained, which is a value comparable with that obtained using KNO3 as nitrogen source and nearly coincident with the theoretical one estimated by the response surface methodology. 相似文献

The effect of ammonium on growth ofCandida apicola and on production of sophorose lipid was studied. Sophorose lipid production increased with increasing initial ammonium sulphate concentration. Both growth and product formation were strongly reduced at 73.6mm ammonium. With 58.9mm ammonium a microcrystalline sophorose lipid was formed. The ratio of the two isomers of the sophorose lipid, harbouring either - or -1 hydroxy fatty acid, was influenced by the initial concentration of ammonium. Both production kinetics, yields and profiles of the total cellular fatty acids express alterations with enlarged ammonium concentrations. The appearance of cytochrome P-450 and of cytochrome oxidase aa3 were determined in the sophorose lipid producing yeast Candida (Torulopsis) apicola IMET 43 747 grown on a mixture of glucose and n-hexadecane. Cytochrome P-450, detectable in both the logarithmic and the stationary growth phase was not repressed by glucose. At the end of the logarithmic growth phase the content of cytochrome P-450 was three- to fivefold increased, which was connected with initiation of sophorose lipid biosynthesis. After that it dropped to the basal level, which remained constant during sophorose lipid biosynthesis. Cytochrome P-450 from logarithmic cells was cross-reactive with an antibody derived against cytochrome P-450alk from C. tropicalis. With microsomal proteins of stationary cells no cross-reactivity was obtained. The microsomal hydroxylase system of stationary cells seem to be regulated by the carbohydrate used as carbon source.

This study examined whether focal adhesion kinase (FAK) plays a role in the differentiation of C(2)C(12) myoblasts into myotubes. Differentiation of C(2)C(12) myoblasts induced by switch to differentiation culture medium was accompanied by a transient reduction of FAK phosphorylation at Tyr-397 (to approximately 50%, at 1 and 2 h), followed by an increase thereafter (to 240% up to 5 days), although FAK protein expression remained unchanged. FAK and phosphorylated FAK were found at the edge of lamellipodia in proliferating cells, whereas the later increase in FAK phosphorylation in differentiating cells was accompanied by its preferential location at the tip of well-organized actin stress fibers. Hyperexpression of FAK autophosphorylation site (Tyr-397) mutant (MT-FAK) reduced FAK phosphorylation at Tyr-397 in proliferating cells and was accompanied by reduction of cyclin D1 and increase of myogenin expression. These cells failed to progress to myotubes in differentiation medium. In contrast, hyperexpression of a wild-type FAK construction (WT-FAK) increased baseline and abolished the transient reduction of FAK phosphorylation at Tyr-397 in serum-starved C(2)C(12) cells. Cells transfected with WT-FAK failed to reduce cyclin D1 and to increase myogenin expression, as well as to progress to terminal differentiation in differentiation medium. These data indicate that FAK signaling plays a critical role in the control of cell cycle as well as in the progression of C(2)C(12) cells to terminal differentiation. Transient inhibition of FAK phosphorylation at Tyr-397 contributes to trigger the myogenic genetic program, but its later activation is also central to terminal differentiation into myotubes. 相似文献

Neutralization of CD95 ligand promotes regeneration and functional recovery after spinal cord injury

The clinical outcome of spinal cord injury (SCI) depends in part on the extent of secondary damage, to which apoptosis contributes. The CD95 and tumor necrosis factor (TNF) ligand/receptor systems play an essential role in various apoptotic mechanisms. To determine the involvement of these ligands in SCI-induced damage, we neutralized the activity of CD95 ligand (CD95L) and/or TNF in spinal cord-injured mice. Therapeutic neutralization of CD95L, but not of TNF, significantly decreased apoptotic cell death after SCI. Mice treated with CD95L-specific antibodies were capable of initiating active hind-limb movements several weeks after injury. The improvement in locomotor performance was mirrored by an increase in regenerating fibers and upregulation of growth-associated protein-43 (GAP-43). Thus, neutralization of CD95L promoted axonal regeneration and functional improvement in injured adult animals. This therapeutic strategy may constitute a potent future treatment for human spinal injury. 相似文献

We studied the association between the honeydew-producing membracid Guayaquila xiphias and its tending ants in the cerrado savanna of Brazil, during 1992 and 1993. Results showed that ants attack potential enemies
of G. xiphias, and that increased ant density near the treehoppers affects the spatial distribution of parasitoid wasps on the host plant,
keeping them away from brood-guarding G. xiphias females. Controlled ant-exclusion experiments revealed that ant presence (seven species) reduces the abundance of G. xiphias’ natural enemies (salticid spiders, syrphid flies, and parasitoid wasps) on the host plant. The data further showed that
ant-tending not only increased homopteran survival, but also conferred a direct reproductive benefit to G. xiphias females, which may abandon the first brood to ants and lay an additional clutch next to the original brood. Two years of
experimental manipulations, however, showed that the degree of protection conferred by tending ants varies yearly, and that
at initially high abundance of natural enemies the ant species differ in their effects on treehopper survival. Ant effects
on treehopper fecundity also varied with time, and with shifts in the abundance of natural enemies. This is the first study
to simultaneously demonstrate conditionality in ant-derived benefits related to both protection and fecundity in an ant-tended
Membracidae, and the first to show the combined action of these effects in the same system.

The Titanosauria were much diversified during the Late Cretaceous, but paleobiological information concerning these sauropods continues to be scarce and no studies have been conducted utilizing modern methods of community analysis to infer possible structural patterns of extinct assemblages. The present study sought to estimate species richness and to investigate the existence of structures in assemblages of the South American Titanosauria during the Late Cretaceous. Estimates of species richness were made utilizing a nonparametric estimator and null models of species co-occurrences and overlapping body sizes were applied to determine the occurrence of structuring in this assemblages. The high estimate of species richness (n = 57) may have been influenced by ecological processes associated with extinction events of sauropod groups and with the structures of the habitats that provided abundant support to the maintenance of large numbers of species. The pseudocommunity analysis did not differ from that expected by chance, indicating the lack of structure in these assemblages. It is possible that these processes originated from phylogenetic inertia, associated with the occurrence of stabilized selection. Additionally, stochastic extinction events and historical factors may also have influenced the formation of the titanosaurian assemblages, in detriment to ecological factors during the Late Cretaceous. However, diagenetic and biostratinomic processes, influenced by the nature of the sedimentary paleoenvironment, could have rendered a random arrangement that would make assemblage structure undetectable. 相似文献
